The included cell line overexpresses ProLink-tagged AGTR1 and EA-tagged β-Arrestin-2. Activation of AGTR1 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. The cAMP Hunter MCHR1 Gi cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. melanin concentrating hormone) based activation of MCHR1 GPCR activity via detection of decreasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type MCHR1, a naturally Gi/Go coupled GPCR. Upon activation of MCHR1, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. In the absence of compound that binds and stabilizes the target, this protein is degraded and no EFC signal is produced. CDC25A is a member of the CDC25 family of phosphatases. CDC25A is required for progression from G1 to the S phase of the cell cycle. It activates the cyclin-dependent kinase CDC2 by removing two phosphate groups. CDC25A is specifically degraded in response to DNA damage, which prevents cells with chromosomal abnormalities from progressing through cell division. CDC25A is an oncogene, although its exact role in oncogenesis has not been demonstrated. Two transcript variants encoding different isoforms have been found for this gene.
